Molecular Analysis of Toxigenic Clostridium difficile Isolates from Hospital Environment by PCR Ribotyping Method

چکیده مقاله:
Background and Aims: Clostridium difficile is an identified cause of antibiotic-associated diarrhea, antibiotic-associated colitis, pseudomembranous colitis and nosocomial diarrhea. The objective of this survey was to determine molecular analysis of toxigenic Clostridium difficile isolates from hospital environment in Tehran tertiary medical centers. Materials and Methods: In this descriptive study, 100 hospital environmental specimens were collected. The specimens were cultured on a selective cycloserine cefoxitin fructose agar, and incubated in anaerobic conditions, at 37°C for 2 days. Clostridium difficile isolates were characterized by conventional biochemical tests. Bacterial cytotoxicity was assayed on tissue culture, and also all strains were typed by PCR ribotyping method. Results: Among toxigenic Clostridium difficile isolates, 6 isolates had the same PCR ribotyping patterns, and 11 isolates were typed in four different groups. Conclusions: Our findings showed that toxigenic Clostridium difficile isolates had different PCR ribotyping patterns. Further studies are recommended to evaluate PCR typing of hospital environmental Clostridium difficile isolates.

A Preliminary Molecular Typing by PCR Assays of Clostridium perfringens and Clostridium difficile Isolates from Dogs
Clostridium perfringens and C. difficile have been associated with acute and chronic large and small bowel diarrhoea, and acute haemorrhagic diarrhoeal syndrome in dogs. The objective of this study was to investigate by toxin gene profile and PCR-ribotyping the molecular characteristics of 14 C. perfringens and 10 C. difficile isolates from 95 canine faeces (n = 36, diarrhoeic and n = 59, non-d...
متن کاملDetection of toxigenic Clostridium difficile in pig feces by PCR
Clostridium difficile is considered an important uncontrolled cause of neonatal diarrhea. Also, the presence of bacteria in the feces of the animal could represent a zoonosic risk for the contamination of meat products. Therefore, it is necessary to have procedures available for the early detection of C. difficile in animals. The current study describes a new semi-automated procedure for the re...
متن کاملThe Frequency of Toxigenic Strains of Clostridium difficile in Hospitalized Patients with Diarrhea in Tehran/Iran by PCR Method, 2010
Background & Aims: Clostridium difficile has been identified as a pathogen in antibiotic associated diarrhea (AAD), pseudomembranous colitis and also nosocomial diarrhea. The present study was performed to find the prevalence of toxigenic strains of C .difficile isolated from diarrhea patients hospitalized in Tehran hospitals. Method: A total of 98 fecal samples obtained during July to December...
متن کاملCharacterization of Clostridium difficile isolates using capillary gel electrophoresis-based PCR ribotyping
We have developed a Clostridium difficile PCR ribotyping method based on capillary gel electrophoresis and have compared it with conventional PCR ribotyping. A total of 146 C. difficile isolates were studied: five isolates were reference strains (PCR ribotypes 001, 014, 017, 027 and 053); 141 were clinical isolates comprising 39 Austrian PCR ribotypes collected in the period 2006-2007 at 25 Aus...
متن کاملMolecular analysis of Clostridium difficile PCR ribotype 027 isolates from Eastern and Western Canada.
The prevalence and characteristics of PCR ribotype 027 strains of Clostridium difficile have come into question following recent outbreaks in Eastern Canada and elsewhere. In order to determine the distribution of this strain in other regions in Canada, we screened a bank of 1,419 isolates recovered from three different Canadian health regions between 2000 and 2004. Among isolates from a Montre...
متن کاملMolecular Evaluation of the Enterotoxigenicity of Clostridium difficile and Clostridium perfringens Swine Isolates by PCR Assays
Clostridium difficile and C. perfringens are enteric pathogens affecting a variety of mammals. This study evaluated the molecular enterotoxigenicity of Clostridium swine isolates by PCRs. One hundred and ten swine faeces were analyzed by culture assay. The faecal samples were from sixty-seven healthy animals and 43 with gastrointestinal tract disease. C. difficile strains were PCR-screened for ...
